珍稀濒危植物北碚榕的组培快繁

摘 要:

以北碚榕茎尖(1.5 cm)为外植体, 通过丛芽诱导、增殖和生根建立离体繁殖体系。结果表明, 茎尖在MS+1.5 mg•L-1 6-BA+0.05 mg•L-1 NAA培养基中培养30 d, 丛芽诱导率达100%, 繁殖系数达9.2; 不定芽在此培养基中继代培养, 1~6代平均繁殖系数达7.0, 丛芽长势良好; 不定芽在1/2MS培养基中, 30 d生根率达100%, 根系发达, 试管苗生长健壮。105株试管苗移栽到土壤营养钵中, 30 d存活102株, 成活率达97.14%。1个茎尖外植体在半年内可繁殖试管苗10万株。

Tissue Culture and Rapid Propagation of Rare and Endangered Ficus beipeiensis

Abstract:

The stem tips (1.5 cm) of Ficus beipeiensis were used as explants to establish a propagation system in vitro. The results indicated that the stem tip explants were cultured on MS medium supplemented with 1.5 mg•L-1 6-BA+0.05 mg•L-1 NAA for 30 days, the induction rate of the adventitious buds achieved 100% and a propagation coefficient reached 9.2. The adventitious buds were subcultured every other 30 day. The Average multiplication coefficient achieved 7.0 from 1 to 6 generations, and the cluster buds grew well. The adventitious buds were trained on 1/2MS basal medium for 30 d, their rooting rate was as high as 100%, the root system developed well and the test-tube plantlets grew vigorously. One hundred and five rooted plantlets were then transplanted to matrix soil, and 102 of them (97.14%) survived after 30 days’ culture. A stem tip can multiply one hundred thousand of test-tube plantlets in six mouths.
